What persistent changes did you make (in /etc/modprobe.d/ for example)? Is the VM set to start automatically? Otherwise just reboot the host, hard if necessary. Turning off IOMMU (in the motherboard BIOS, of via the bootloader edit menu) can help prevent VMs with PCIe passthrough to start.
