After the VM is built, is it advise that you unmount the cloud-init drive? I assume on the next reboot it will attempt to re-initialize the VM? Am i correct to assume that?

It's supposed to stay there. If you didn't disable cloudinit inside the VM, it won't even boot.
I don't understand what you mean.

I'm going through a lab trying to setup a template for my debian hosts. I followed some instructions, every thing works as expected and perfectly. My question was if the cloudinit should get unmounted.

If you say it should stay there, whats the reason? I unmounted it and it booted fine.

Hm, well then. I have a Ubuntu cloudinit template and its boot sequence relies on the cloudinit disk. If it can't find it then the boot sequence doesn't finish.
But if it works without, why are you even asking? :)

Just trying to understand, thats all. If it shouldn't boot without it then maybe i goofed. I'll try again.
