If I were to setup a VM using local storage, could I then move that VM to Ceph storage at a later time?

Yes. No problem if both storages are on this server. Just Hardware Tab -> Disk move (VM should be offline)

If that does not work or it is not on the same server, do a backup and restore, which will pop up a storage destination window.

I am actually hoping to build a VM on a temporary PVE host using local storage, then move the VM to a new cluster that will be using Ceph.

VM don't need to be offline ... you can move disk from one storage to another of a running VM with no downtime ... that's black magic to me but it works like a charm !
