I have a DL380p and the backplane is connected to an H220 HBA card.

Now I should choose a reliable way to add two 2.5" SSD to put in ZFS mirror to boot proxmox. How bad is to connect one to optical SATA port and the other one to a USB to SATA adapter? (USB port is internal and 2.0)

Another solution is to add a PCIe SATA card but, if possibile, I'd like to keep free a slot for further expansions (the other 5 are already filled).
 
Probably not that good of an idea, especially since the USB port is 2.0 . USB to SATA should be performing a lot worse than SATA, since USB is not made primarily for connecting disks. Even with 3.0 it still would be a bad idea, since you get worse performance.

"optical SATA port"? I have used a USB 2.0 to SATA converter for an optical disc drive and it was fast enough for Blu-ray, but I don't recommend it for SSD.

optical because its purpose (officially) is to attach an optical drive, but it is a full featured SATA3 port.

Yes, it's a no no solution. I searched and the 2.0 has 500 mA capability. Too less even for a SSD (for mine sure: it has 5W under load)
