Hello, I need help guys. I have a separate 500GB HDD drive, I created it as a Directory through the GUI in ext4, created one partition /dev/sdc1, but did not find how to make a second partition /dev/sdc2 in the GUI.
Is it possible to split a 250GB Directory disk into 2 partitions and how to do it?

Delete the Directory storage from PVE GUI, under Node/Disks wipe the relevant disk. Follow one of many guides on the net, ie:
https://gcore.com/learning/how-to-partition-a-disk-in-linux/

make sure you understand the steps and apply them to the correct disk

good luck

thanks for the answer, try to do it tonight. I have already tried to do something like this guide, but a little different, but my proxmox did not start and went into emergency mode due to entries in /etc/fstab

If you created a manual entry in fstab, you want to unmount the disk (after you delete the Directory storage) and remove that entry, before you wipe the disk.

everything turned out to be done, but this storage is not shown on the left so that I can use it for backups. Is it possible to do this?

There is not enough information to say "yes" or "no".

Theoretically it is possible, if you are in a state I suspect you are, then you still need to format the partitions, mount them, add directory type storage into PVE and mark it for an approriate content (backup).
